For 3.5" and 5.25", which of the following alignment disks partnumber would you recommend I get please?

I am using the AAD 206-34, P/N: 810251A
I am also using the Dysan 224 alignment disk.

No, I have found that 3.5" drives don't go out of alignment, and if they were I would throw it away and buy another one.
